Prepare cells
1. Check and confirm that the cells are not contaminated with fungi, bacteria, or mycoplasma.
2. The cells before cryopreservation are best in the logarithmic growth phase of vigorous proliferation.
3. Count the number of cells to be frozen. Suspended cells should be centrifuged at 180g for 5-10 minutes before removing the culture medium. Adherent cells should be digested and counted before centrifugation at 180g to obtain cell pellet.
Cell cryopreservation
1. Firstly, determine the appropriate freezing density and volume for cells, and calculate the volume of LC1601 freezing solution that should be used. [Note]: The usable freezing density is 1-10 × 106/mL, and the recommended freezing density is 2-5 × 106/mL. The freezing volume is 5-1 mL/tube.
2. Take an appropriate amount of rapid cell cryopreservation solution (serum-free, protein free) (item number: AC05L033) and resuspend the cells. The product should be pre cooled at 4 ℃ before use. After cell resuspension, it should be immediately divided into cell cryovials.
3. Cells resuspended in frozen storage solution should not be allowed to stand at room temperature or 4 ℃ for more than 10 minutes. The frozen tube should be transferred to a -80 ℃ freezer as soon as possible, and no programmed cooling measures or devices should be taken. If it is not possible to immediately move into a -80 ℃ low-temperature refrigerator, at least it should be temporarily stored in an environment below 0 ℃ (such as a -20 ℃ refrigerator) for about 10 minutes, and no program cooling should be taken.
4. After overnight storage in a -80 ℃ freezer, if stored for a long time, it can be transferred to a liquid nitrogen tank for preservation. If used in the short term, it can be stored at -80 ℃ for several months, and the storage time and effect vary depending on the cell type. [Note]: (1) The cell types frozen for the first time using this product should be tested for storage time and effectiveness before use. (2) Reference rapid cell cryopreservation solution (serum-free, protein free) (item number: AC05L033), stored at -80 ℃ for 12 months at a rate of 1 × 106/mL per tube, and revived with an immediate viability rate of>90%.
Cell revival
1. Prepare a complete culture medium pre warmed at room temperature or 37 ℃, with a volume 9 times that of the frozen storage solution.
2. Remove the cryovial from liquid nitrogen or -80 ℃, place it in a 37 ℃ water bath and shake gently continuously. When there is one grain of ice left, quickly remove it from the water bath.
3. Immediately transfer the frozen solution to the complete culture medium, gently blow and mix 3-5 times. Then, centrifuge 180g for 5-10 minutes to obtain cell pellet for later use.
