Transportation and storage
Transport at room temperature. Store at room temperature and avoid light, with a shelf life of 12 months.
Usage
1. For adherent cells (using a 6-well plate as an example)
(1) Cultivate cells using a 6-well plate.
(2) Place the 6-well plate on ice and rinse the cells twice with pre cooled 4 ℃ PBS.
(3) Fix cells with pre cooled 4 ℃ 100% methanol for 10 minutes. Ensure complete coverage of cells with methanol to achieve uniform fixation effect.
(4) Extract methanol from the well plate, and if the experiment needs to be temporarily stopped, cover the cells with PBS containing 50% glycerol. Wrap the 6-well plate with sealing film or cling film and store it in a refrigerator at 4 ℃ for one week.
(5) Remove the cells from the ice and allow them to equilibrate at room temperature. Cover the cells with an appropriate amount of crystal violet staining solution and incubate at room temperature for 10 minutes. Discard the crystal violet staining solution, which can be reused.
(6) Rinse the cells multiple times with distilled water until there are no more residual crystal violet in the washing solution. Ensure the removal of excess dye and avoid background interference.
(7) Microscopic examination.
