Transportation and storage
Transport at room temperature. Component A is stored at 4 ℃, while component B is stored at room temperature; Valid for 24 months.
Usage
1. Preparation work
(1) Preparation of M-Buffer EX Column: After receiving the reagent kit, take out the M-Buffer EX Column centrifuge column and mix it upside down for about 10 times (ensuring that the white medium is fully mixed), then store it upright and let it stand. Note: If the reagent kit is received, it should be used immediately and left upright for 2-5 minutes before use.
(2) Assembly: Tear off the sealing film on the top cover of the centrifuge column, remove the top cover, then remove the bottom cover and place it into the Collection Tubes (15mL) collection tube.
(3) Remove protective solution: 1000g, centrifuge for 2 minutes, remove the centrifuge column, discard the protective solution from the collection tube, and place the centrifuge column back into the recovery collection tube.
2. M-Buffer EX Column Balance
(1) Add 2.4mL of equilibrium buffer (sample buffer to be replaced) to M-Buffer EX Column, at 4 ℃, 1000g, centrifuge for 2 minutes, and discard the waste liquid in the collection tube.
(2) Repeat step 2 (1) to replace the centrifuge column with a new 15mL centrifuge tube. Note: Do not discard the original 15mL Collection Tubes. This collection tube can be used specifically for waste liquid collection or for upright storage of M-Buffer EX columns.
3. Buffer replacement/desalination/removal of small molecules
(1) Load sample: Slowly add up to 0.5mL of sample to the center position of M-Buffer EX Column. Note: The recommended sample volume is 0.2-0.5mL; For samples with a volume less than 0.2mL, the sample can be diluted to 0.2mL or a smaller volume buffer displacement device can be selected; For samples with a volume greater than 0.5mL, they can be divided into multiple samples for operation or a larger volume buffer replacement device can be selected.
(2) Buffer replacement: 4 ℃, 1000g, centrifuge for 2 minutes, remove the centrifuge column, collect 15mL of liquid in the centrifuge tube, complete buffer replacement, and can be used for the next experiment.
4. M-Buffer EX Column cleaning and storage
(1) Cleaning: After completing the buffer replacement, add 2.4mL of deionized water, 1000g, centrifuge for 2 minutes, and discard the waste liquid in the collection tube into the M-Buffer EX Column.
(2) Repeat cleaning twice.
(3) Storage: Add 2.4mL of 20% ethanol, 1000g, centrifuge for 2 minutes, and discard the waste liquid in the collection tube. Finally, fill the M-Buffer EX Column with 20% ethanol, block the lower cover first, and then close the top cover. Mix the mixture upside down for about 10 times (ensuring that the white medium is fully mixed), and store it upright at 4 ℃. Note: M-Buffer EX Column is recommended to be reused 3-5 times before replacing with a new product, otherwise it will affect the buffer replacement effect of the centrifugal column.
