The live cell apoptosis detection kit is based on a fluorescent inhibitor of caspase. These inhibitors have cell permeability and non cytotoxicity. Once inside the cell, caspase inhibitors covalently bind with active caspase. Caspase-1 is mainly involved in the activation of pro-inflammatory cytokines and the process of cell apoptosis. It has been demonstrated that caspase 1 exhibits substrate selectivity towards the peptide sequence Tyr Val Ala Asp (YVAD). This kit uses FAM-YVAD-FMK as a fluorescent indicator for caspase 1 activity. FAM-YVAD-FMK irreversibly binds to activated caspase 1 in apoptotic cells. Once bound to cysteine protease 1, the fluorescent reagent remains within the cell. There are multiple parameters that can be used to monitor cell apoptosis by combining inhibition of caspase-1 without preventing apoptosis.
