Wheat germ agglutinin, labeled AF555 (hereinafter referred to as "AF555 WAG"), is a lectin that binds to N-acetyl-D-glucosamine and sialic acid, and may be the brightest WGA conjugate in bright and red fluorescence. It is currently a widely studied and applied type of lectin. WGA binds to glycoconjugates, and its derivatives and conjugates are widely used for labeling cell membranes and fibrotic scar tissues for fluorescence imaging and analysis. The sugar binding specificity of WGA is related to the sequence of β -1,4-GlcNAc linking residues, i.e. chitinase. Each monomer contains two identical non interacting binding sites that complement 3 or 4 β -1,4-GlcNAc units. Among the detected monosaccharides, only GlcNAc binds to WGA. Mannan does not bind, while GalNAc can only weakly bind. WGA has a high affinity for binding to internal GlcNAc residues in large oligosaccharides containing Gal β (1-4) GlcNAc β (1-3) repeat sequences (i.e., galactosaminoglycans). N-acetylneuraminic acid only participates in low affinity interactions with WGA. WGA displays complex carbohydrate specific patterns that can be used for structural analysis of complex carbohydrates.
