Product Description
LyGreen nucleic acid dye is a novel double stranded DNA binding saturated fluorescent dye, which is a new generation of fluorescent dye used for HRM analysis and can also be used for real-time quantitative PCR (qPCR). The dye structure consists of two monomeric dye molecules connected by an elastic arm to form a dimer. In the absence of DNA, the structure forms a non fluorescent circular structure. Once DNA appears, it returns to its random form and binds to DNA, emitting strong green fluorescence.
Compared with other fluorescent dyes on the market such as SYBR Green I, LyGreen has the following advantages: (1) high sensitivity: it can obtain a stronger signal than SYBR Green I when used at the recommended concentration; (2) accuracy: it binds to double stranded DNA, and the dye does not migrate during deformation, and the fluorescence signal accurately reflects the changes in double stranded DNA. Excessive dye forms dimers without dissociation, resulting in much lower inhibition of PCR by LyGreen compared to SYBR Green I. Therefore, qPCR experiments using LyGreen can use rapid PCR steps. (3) Good stability: It will not be damaged during normal storage, operation, and PCR processes. Dyes in buffer solutions can be safely stored at room temperature or in the refrigerator, and can also be repeatedly frozen and thawed. (4) Safety: Cell membrane penetration testing shows that LyGreen can hardly penetrate the cell membrane. (5) Compatibility: Similar to SYBR Green I spectrum, there is no need to change the spectral element settings or operating steps when using different brands of qPCR instruments. After qPCR is completed, it can be directly observed and analyzed in the gel imaging system without adding other nucleic acid dyes. (6) Good repeatability (as shown in Figure 1): Specially designed dyes, under the same reaction system, the fluorescence signal release is only related to the double chain concentration, with perfect linearity.
