Product Description

LiFluor 488 succinimide ester is one of the LiFluor series fluorescent labeling dyes that can cover the entire visible spectrum. All LiFluor dyes have excellent water solubility, and their hydrophilicity minimizes the use of organic solvents. Compared with conventional dyes such as FITC, TRITC, Texas Red, Cy3, Cy5, and Cy7, LiFluor dye has better labeling performance. Some LiFluor dyes are significantly superior to Alexa Fluor labeling dyes on certain antibodies. They are extremely inexpensive fluorescent dyes used for labeling proteins and nucleic acids (replacing Alexa Fluor dyes). The development of each LiFluor dye is matched with the spectral characteristics of specific Alexa Fluor or other marker dyes (such as DyLight dye).

Succinimide based (NHS) esters have been proven to be reagents for amine modification, as the formed amide bonds are essentially identical and stable to natural peptide bonds. These reagents are usually stable and exhibit good reactivity and selectivity with aliphatic amines. When succinimide ester compounds are used for conjugation reactions, there are few factors to consider: 1. Solvent: In most cases, the reactive dye should be dissolved in anhydrous dimethylformamide (DMF) or dimethyl sulfoxide (DMSO). 2. Reaction pH: The labeling reaction between amine and succinimide ester strongly depends on pH. Amine reactive reagents react with non protonated aliphatic amine groups, including the terminal amine of proteins and the beta amino group of lysine. Therefore, the aminoacyl reaction usually occurs at pH 7.5 or above. Proteins modified with succinimide esters are representative and are typically synthesized at pH 8.5-9.5. 3. Reaction buffer: When using amine reaction reagents, it is necessary to avoid using buffer solutions containing free amines (such as Tris and glycine) and thiol compounds. Ammonium salts widely used for protein precipitation, such as ammonium sulfate and ammonium acetate, must also be removed before dye conjugation (e.g. by dialysis). 4. Reaction temperature: Most conjugations are carried out at room temperature. Specific labeling reactions may require an increase or decrease in temperature. LiFluor series dyes are substitutes for AF series dyes.
