Prepare cells
1. Check and confirm that the cells are not contaminated with fungi, bacteria, or mycoplasma.
2. The cells before cryopreservation are best in the logarithmic growth phase of vigorous proliferation.
3. Count the number of cells to be frozen. Suspended cells should be centrifuged for 5-10 minutes before removing the culture medium, while adherent cells should be digested and counted before centrifugation to remove the culture medium and obtain cell pellet.
Cell cryopreservation
1. The cell cryopreservation solution should be pre cooled at 4 ℃ before use.
2. Determine the appropriate freezing density and volume for cells, calculate the volume of freezing solution to be used, and recommend a freezing density of 2 * 106/mL-1 * 107/mL.
3. Take an appropriate amount of frozen solution and resuspend the cell pellet to prepare a suspension. The cell suspension should be immediately divided into cell freezing tubes. Cell suspensions prepared using cryoprotectants should not be left at room temperature or 4 ℃ for more than 10 minutes.
5. Move the cryovial into a -80 ℃ freezer and let it sit overnight without the need for programmed cooling.
6. Transfer the cryovials stored overnight at -80 ℃ into liquid nitrogen for preservation. If used for a short period of time, it can be stored at -80 ℃, and the storage time and effect vary depending on the cell type.
Cell revival
1. Pre warm the complete cell culture medium at room temperature or 37 ℃, with a volume 9 times that of the frozen solution.

3. Suck the cell suspension into complete culture medium, gently mix well, centrifuge, remove the supernatant, and obtain a cell precipitate for later use.
