Mycoplasma is a prokaryotic organism without a cell wall, measuring approximately 0.1-0.6 micrometers in size. Currently, over 100 species of mycoplasma have been discovered. Due to the small diameter of mycoplasma, it can often pass through the laboratory's conventional 0.20 micron pore size sterilization membrane, and even pass through 0.10 micron pore size sterilization membrane during high-pressure filtration, making it difficult to remove mycoplasma contamination from cells.
The mycoplasma clearance reagent I developed by our company contains drugs that inhibit the synthesis of mycoplasma proteins, while the mycoplasma clearance reagent II contains drugs that inhibit the synthesis of mycoplasma DNA. Mycoplasma clearance reagent I requires at least 15 days of treatment, while mycoplasma clearance reagent II only requires 7 days of treatment. Both can achieve the goal of permanently killing and eliminating mycoplasma.
About 5-10% of mycoplasma will develop resistance to Quick Cell Mycoplasma Elimination Reagent I or Quick Cell Mycoplasma Elimination Reagent II. In addition, about 3-5% of cells will die during the process of killing mycoplasma due to the high toxicity of the drug to the cells. Due to the above two reasons, we generally recommend dividing contaminated cells into two parts and using two different reagents to kill them separately. This way, the probability of mycoplasma developing resistance to both drugs and cells dying simultaneously during treatment will be very low, allowing us to select the most effective removal reagent. For cases where no single reagent can completely kill Mycoplasma, we also recommend using a combination of two removal reagents at the same time. In this case, it can basically ensure the complete removal of Mycoplasma, but the toxicity of both reagents to cells will be greater, and cell conservation work needs to be done in advance.
