The molecular formula of CFDA-SE is C29H19NO11, with a molecular weight of 557.47 and a CAS number of 150347-59-4. The English name of CFDA-SE is carboxyfluorescein diacetate and succinimide ester. CFDA-SE can be used for cell tracking and cell proliferation detection. CFDA SE can be broken down into CFSE by intracellular esterases after entering cells through intact cell membranes. CFSE can randomly and irreversibly bind to lysine residues or other amino groups in intracellular proteins and label these proteins. After adding the fluorescent probe CFDA SE, cells can be fully labeled approximately 24 hours later. The fluorescence of non dividing cells labeled with CFDA SE is very stable, and the labeling time can reach several months.
CFDA-SE labeled cells exhibit green fluorescence, and the excitation wavelength for detection can be selected as 488 nm excitation, with an emission wavelength of 518 nm. FL1 detection channel can be used for flow cytometry detection. In addition to detecting cell proliferation using flow cytometry, the number of live cells can also be quantified using a fluorescent enzyme-linked immunosorbent assay (ELISA) plate, or uniformly stained cells can be observed using a fluorescence microscope for cell tracking.
The fluorescence of the progeny cells labeled with CFDA-SE decreases by half every time they divide, so that non dividing cells can be detected by flow cytometry. The fluorescence intensity of cells that divide once is halved (1/2), the fluorescence intensity of cells that separate twice is halved (1/4), and so on. The fluorescence intensity gradually decreases according to the rules of 1/2, 1/4, 1/8, and 1/16. The detection results obtained by flow cytometry using CFDA-SE indicate that each peak represents a cell with a certain number of divisions. The peaks from right to left are usually cells with 0, 1, 2, 3 divisions, and other numbers of divisions. After a high number of divisions, cells with zero or one division or fewer divisions will gradually decrease until they cannot be detected. CFSA-SE can detect up to eight or even more splits. Cells labeled with CFDA-SE can be used for in vitro and in vivo proliferation studies, and have the function of not staining adjacent cells. CFDA-SE is most commonly used for lymphocyte proliferation detection, and can also be used for proliferation detection of other cells such as fibroblasts, natural killer cells, hematopoietic progenitor cells, etc. If CFDA-SE is used in combination with other fluorescent probes, it can obtain other relevant information of cells with different division times.
