Cell proliferation is an important indicator for evaluating cell activity, metabolism, physiological and pathological conditions. EdU (5-Ethynyl-2 ` - deoxyuridine) is a thymidine analogue with alkyne groups that are rare in natural compounds. It can replace thymidine (T) and be incorporated into replicating DNA molecules during cell proliferation. Through a specific conjugation reaction between EdU and fluorescent dyes, the fluorescent groups are labeled onto newly synthesized DNA containing EdU, allowing for efficient and rapid detection of DNA replication activity. This technology is widely used in research on cell proliferation, cell differentiation, growth and development, DNA damage repair, and other fields.
The traditional immunofluorescence staining (BrdU) detection method requires complex and tedious operational steps such as DNA denaturation, antigen repair, and overnight incubation of antigen antibodies. The EdU detection method does not require severe DNA denaturation, only mild cell immobilization and permeabilization treatment, which can better protect cell morphology, overall DNA structure, and intracellular antigen recognition sites. The operation steps are faster, more sensitive, and accurate. EdU has a very similar structure to thymine (T), but its size is only 1/500 of BrdU antibody, making it easy to spread within cells; No antigen antibody reaction is required, which can quickly and conveniently reflect DNA replication activity at the cellular and tissue levels.
