Hoechst 33258 is a blue fluorescent dye that can penetrate the cell membrane and has low toxicity to cells. It releases strong blue fluorescence after embedding into double stranded DNA. Hoechst 33258 is a specific DNA dye that binds to A-T bonds. This dye can immediately stain dead cells or cells fixed in 70% cold ethanol. The staining of living cells is gradual and can reach saturation within 10 minutes. Under a fluorescence microscope, live cell nuclei exhibit diffuse and uniform fluorescence. When apoptosis occurs, dense granular fluorescence can be seen in the nucleus or cytoplasm. Hoechst 33258 is commonly used for detecting cell apoptosis, which can be observed under a fluorescence microscope or detected by flow cytometry after staining. It can also be used for imaging observation of ordinary cell nuclei and mitochondria.
Hoechst is often used as a substitute for other nucleic acid dyes such as DAPI. The key difference between these two dyes is that Hoechst 33258 has stronger lipophilicity compared to DAPI, making it better able to penetrate intact cell membranes. The recommended working concentration of Hoechst 33258 for nuclear staining is 0.5-10 μ g/mL.
