There are three commonly used enzymes in digestion: trypsin, collagenase, and hyaluronidase. Hyaluronidase is often used in combination with trypsin or collagenase. Trypsin has a strong effect and is prone to damage smooth muscle cells, while collagenase has a milder effect and can digest collagen fibers in the intercellular matrix to release cells, causing minimal damage to cells. It is the only protein enzyme that can degrade natural collagen fibers with a triple helix structure.
Collagenase is a crude enzyme extract extracted from Clostridium histolyticum, also known as Clostridium peptidase A. It contains not only collagenase, but also other proteases, polysaccharides, lipases, etc. These components can effectively hydrolyze other proteins, polysaccharides, and lipids in the extracellular matrix of connective tissue and epithelial tissue, respectively. The enzyme has good separation effect and remains active even in the presence of calcium and magnesium ions. Therefore, it can be prepared with PBS and serum containing culture medium, which is easy to operate and can improve cell survival rate. The final concentration is 200u/mL. The digestion effect of this enzyme is gentle and does not require mechanical oscillation.
At present, commercially available bacterial collagenase is mainly divided into four types based on differences in collagenase activity: collagenase type I, type II, type III, and type IV, with some bias in application: (1) Type I collagenase: contains relatively uniform enzyme activity (including collagenase, caseinase, clostridian protease, trypsin activity). Usually used for the preparation of epithelial cells, liver, lung, adipose and adrenal tissue cells. (2) Type II collagenase: contains higher activity of clostridial protease and is commonly used for the preparation of tissue derived cells such as heart, bone, muscle, thymus, and cartilage. (3) Type III Collagenase: Contains low protease activity and is commonly used in the preparation of breast cells. (4) Type IV Collagenase: Contains low trypsin activity and is commonly used for the preparation of pancreatic islet cells or cell preparation experiments that require maintaining receptor integrity.
