The decrease in mitochondrial membrane potential is a sign of early apoptosis in cells, which occurs before the outward rotation of phosphatidylserine and activation of caspase hydrolase on the cell membrane. When the permeability of mitochondrial membrane changes, the membrane potential will decrease. The change in membrane potential is caused by the formation of Bax dimer and the activation of Bid, Bak, and Bad, which induce the formation of pores in the mitochondrial membrane. When these pro apoptotic proteins are activated, mitochondria also release cytochrome C into the cytoplasm.
JC-1 is an ideal fluorescent probe widely used for detecting mitochondrial membrane potential (Δ PSI m), which can detect mitochondrial membrane potential in cells, tissues, or purified samples. When the mitochondrial membrane potential is high, JC-1 aggregates in the mitochondrial matrix, forming a polymer that produces red fluorescence; When the mitochondrial membrane potential is low, JC-1 cannot aggregate in the mitochondrial matrix. At this time, JC-1 is a monomer and can produce green fluorescence. The transition of JC-1 from red fluorescence to green fluorescence can easily detect the decrease in membrane potential, and the transition of JC-1 from red fluorescence to green fluorescence can also be used as an early detection indicator of cell apoptosis.
The maximum excitation wavelength of JC-1 monomer is 510 nm, and the maximum emission wavelength is 527 nm; The maximum excitation wavelength of JC-1 polymer is 585 nm, and the maximum emission wavelength is 590 nm. This reagent kit is easy and fast to operate, and can be detected by flow cytometry, fluorescence microscopy, or fluorescence enzyme-linked immunosorbent assay.
