Product Description

The Cell Cycle and Apoptosis Analysis Kit is a detection kit that uses the classic Propidium iodide staining (PI staining) method for cell cycle and apoptosis analysis.

Propidium iodide (PI) is a fluorescent dye for double stranded DNA. The combination of propidium iodide and double stranded DNA can produce fluorescence, and the fluorescence intensity is proportional to the content of double stranded DNA. After the DNA inside the cell is stained with propidium iodide, the DNA content of the cell can be measured using a flow cytometer. Based on the distribution of DNA content, cell cycle and apoptosis analysis can be performed.

After staining with propidium iodide, assuming the fluorescence intensity of G0/G1 phase cells is 1, the theoretical value of the fluorescence intensity of G2/M phase cells containing duplicated genomic DNA is 2, and the fluorescence intensity of S phase cells undergoing DNA replication is between 1-2. Apoptotic cells lose some genomic DNA fragments during the staining process due to nuclear condensation and DNA fragmentation. Therefore, after staining with propidium iodide, apoptotic cells show obvious weak staining, with a fluorescence intensity less than 1. The so-called sub-G1 peak, also known as the apoptotic cell peak, appears on the flow cytometry fluorescence image.

When cells undergo apoptosis, apoptotic bodies are produced due to the concentration of cytoplasm and chromatin, as well as nuclear fragmentation, which changes the light scattering properties of the cells. In the early stages of cell apoptosis, the ability of cells to scatter forward angular light significantly decreases, while their ability to scatter lateral light increases or remains unchanged. In the late stage of cell apoptosis, both forward and lateral light scattering signals decrease. Therefore, changes in cell light scattering can be measured by flow cytometry to observe cell apoptosis.

This kit is typically used for detecting cell cycle and apoptosis in cultured adherent or suspended cells. If used for cell cycle and apoptosis detection in tissues, the tissue must be digested into a single-cell state before detection can be performed.
