Transportation and storage
Transport at room temperature. Stored at 4 ℃, packaged with ammonium persulfate solution and stored at -20 ℃, with a shelf life of 12 months.
Instructions for use(refer to the diagram at the end of the article)
1. Preparation of reagents
(1) Before the first use, take 5ml of deionized water and add it to an ammonium persulfate (APS) test tube, shake it appropriately, mix well, and place it at 4 ℃ for later use. It is recommended to store it at -20 ℃ for long-term use.
(2) According to the adhesive mold used in the experiment, take 1/2 volume of separation adhesive A and separation adhesive B respectively, and add them to a small beaker (test tube) to mix well for later use. [Example]: When using a Bole Mini PROTEAN electrophoresis tank to prepare gel, take 2.0/2.5/3.8 ml of gel A and B from each 0.75/1.0/1.5 mm gel.
(3) Take another small beaker and take 1/2 volume of concentrated gel solution A and concentrated gel solution B separately. Mix well and set aside for later use. [Example]: When using the Bole Mini PROTEAN electrophoresis tank, for each 0.75/1.0/1.5mm gel, 0.8/1.0/1.5ml of concentrated gel A solution and concentrated gel B solution are taken respectively.
(4) Add 10% APS solution (that is, add 10 μ l 10% APS solution to 1ml gel mixture) into the separation gel mixture and concentrated gel mixture prepared in the first two steps at a ratio of 1:100, and gently mix them to avoid bubbles.
2. Glue filling operation
(1) Pour an appropriate amount of the separation gel mixture prepared in the previous step into the gel mold (for the use of Bole Mini PROTEAN electrophoresis tank, add the gel solution to about 1.5 cm from the top of the front glass plate or about 0.5 cm from the comb teeth); There is no need to wait and immediately pour the concentrated adhesive mixture prepared in the previous step into the glass plate.
(2) Insert the comb into the gel, let it stand for 8-15 min, and wait for gel polymerization. [Note]: The prepared gel can be stored at 4 ℃ for several weeks in a sealed bag with a small amount of electrophoresis buffer.
3. Electrophoretic operation
(1) After the gel is polymerized, assemble the electrophoresis tank, add the electrophoresis buffer, carefully pull out the comb, check the glue hole and tidy the comb teeth.
(2) Add protein samples and protein molecular weight standards as needed for the experiment.
(3) Adjust the electrophoresis instrument to a constant voltage of 110V-150V for electrophoresis operation. [Note]: If the current of the single board adhesive is greater than 90mA and the current of the double board adhesive is greater than 130mA, the voltage can be appropriately reduced.
matters needing attention
1. This product is only for scientific experimental research and should not be used in clinical diagnosis, treatment, or other fields.
2. This kit has essential differences from traditional SDS-PAGE. Please follow the product manual for operation. If you have any questions, you can refer to the product operation video file provided by our company or contact our technical personnel.
3. The prepared ammonium persulfate should be stored at -20 ℃. If stored at 4 ℃, it can only be used for one week.
4. SDS has been added to the separation buffer and concentration buffer of this reagent kit.
