Product Description
Protease K, with a relative molecular weight of approximately 29.3 kDa, is a serine protease that can cleave the carboxyl terminal peptide bonds of both aliphatic and aromatic amino acids. It has a wide range of cleavage activities. It has activity in a wide pH range (pH 4-12.5). The general working concentration is 50-100 µ g/mL, but it can enhance activity in denaturing agents such as 1% SDS. Therefore, the specific working concentration is often determined based on factors such as SDS, urea, pH, temperature, etc. in the buffer solution used during use. Common concentrations of EDTA Triton X-100、Tween 20、Sarkosyl、 Guanidine hydrochloride has little effect on the activity of protease K.
Commonly used for experiments such as genome DNA extraction and preparation of chromosomal DNA by pulse electrophoresis.

usage method
1. Preparation of proteinase K solution: Generally, a proteinase K solution with a concentration of 20 mg/mL is prepared. For example, 200 mg of proteinase K can be added to 9.5 ml of dd water and gently shaken until proteinase K is completely dissolved. Do not vortex and mix, add water to make up to 10 mL, and finally divide into small portions and store at -20 ℃.
2. According to the final concentration of 50-100 µ g/mL, take an appropriate amount of proteinase K solution and add it to the sample to be digested. Incubate at a temperature of 55-65 ℃, with an ideal incubation temperature of 58 ℃. Incubate for 15 minutes to 48 hours, depending on the sample situation.

1. This product is only for scientific experimental research and should not be used in clinical diagnosis, treatment, or other fields.
2. If 20 mg/mL of proteinase K needs to be further diluted, it is recommended to use 10 mM Tris HCl at pH 7.4~7.8 for dilution.
