Hygromycin B is an aminoglycoside antibiotic produced by the metabolism of Streptomyces hygroscopicus. It inhibits protein synthesis by interfering with 70S ribosome translocation and inducing misreading of mRNA templates, thereby killing prokaryotic (such as bacteria), eukaryotic (such as yeast, fungi), and higher mammalian eukaryotic cells.
The hygromycin resistance gene (hyg or hph) derived from Escherichia coli encodes the hygromycin B phosphotransferase, which converts hygromycin B into biologically inactive phosphorylated products, thereby playing a detoxifying role. Based on this principle, hygromycin B is a very useful selective marker used to screen and maintain prokaryotic or eukaryotic cells successfully transfected with hygromycin resistance genes in culture. In addition, due to differences in the mode of action, it is often used in combination with G418 (Cat: B11L234), Zeocin (Cat: B11L251), and Blasticidin S (Cat: B11L221) for the selection of dual resistance positive cell lines. Chloramphenicol B can also be used as an antiviral agent, as it selectively penetrates into cells with increased permeability due to viral infection and has the effect of inhibiting translation. It can also be mixed into animal feed to provide deworming function.
