Product Description
Blasticidin S is a nucleoside antibiotic derived from Streptomyces griseochromenes, commonly known in Chinese as blasticidin S, rice blast fungicide S, or rice blast powder. It is usually a hydrochloride salt. Blasticidin S is commonly used to screen stable mammalian transfected cell lines carrying bsr/BSD/bls gene (commonly labeled as bsrr/bsdr/Blastr) plasmids. It has a rapid and potent mode of action, and low antibiotic concentrations can cause rapid cell death in cells without resistance genes.
This product is packaged and prepared in HEPES (pH 7.4) buffer solution at a concentration of 10mg/ml. It is filtered through a 0.22 μ m filter membrane for sterilization and can be directly used for cell culture. The 10mg and 50mg packaging of this product are in powder form.
Blasticidin S specifically inhibits protein synthesis in prokaryotic or eukaryotic cells by interfering with ribosome binding peptide segments. There are currently three types of blast resistant genes, one of which is the acetyltransferase gene bls isolated from Streptomyces sp; The other two are deaminase genes, namely bsr isolated from Bacillus cereus and BSD gene isolated from Aspergillus terreus. The bsr and BSD genes are the most commonly used screening markers for stable cell line screening in mammalian and plant cells. Blasticidin S can also be used for screening prokaryotic cells such as Escherichia coli.
Blasticidin S has a rapid and potent mode of action, which can cause rapid cell death at very low antibiotic concentrations. Escherichia coli is typically sensitive to concentrations of 50 µ g/ml, while mammalian cells are sensitive to concentrations as low as 2-10 µ g/ml. Cell death occurs rapidly and can typically form stable mammalian cell lines with Blasticidin S resistance in less than a week.
