Product Description
Puromycin is an amino nucleoside antibiotic derived from Streptomyces alboniger, also known as puromycin in Chinese. It is commonly used to screen eukaryotic or prokaryotic polyclonal or monoclonal cells that can express the pac gene (puror) through plasmid transfection/transformation, viral infection, and other methods. Puromycin is not only used for screening stable cell lines, but also for maintaining stable cell lines. The 10mg/ml packaging of this product has been filtered and sterilized, and can be directly used for cell culture.
The characteristic of Puromycin is its rapid action on cells, generally killing 99% of cells that do not express the pac gene within 2 days.
In Gram positive bacteria, animals, or insect cells, puromycin inhibits or kills cells by suppressing protein synthesis. Its mechanism of action is that puromycin is an analogue of the aminoacyl tRNA molecule at the 3 'end, which can bind to the A site of the ribosome and be incorporated into the extended peptide chain. After binding to the A site, puromycin does not participate in any subsequent reactions, leading to premature termination of protein synthesis and the release of immature peptides containing puromycin at the C-terminus.
The Pac gene expresses Puromycin N-acetyl-transferase, which is found in Streptomyces alboniger. If the gene is expressed, it will develop resistance to puromycin, which is currently widely used in screening stable mammalian cell lines expressing the pac gene. For example, many commercial lentiviral vectors carry the pac gene (usually labeled as puror on plasmid maps), allowing for the screening of stable expression cell lines of specific genes using puromycin. Purimycin can also be used to screen Escherichia coli strains, yeast strains, etc. that express the pac gene.
