Zeocin, also known as phytomycin D1 or bleomycin D1 in Chinese, is a member of the bleomycin/phytomycin antibiotic family isolated from a mutant strain of Streptomyces verticillus. It has inhibitory effects and cytotoxicity on bacteria, fungi (including yeast), plants, and mammalian cells.
Zeocin is an alkaline, water-soluble, copper ion chelating glycopeptide antibiotic. The Zeocin solution chelated with Cu2+appears blue and inactive. When it enters the cell, Cu2+on Zeocin is reduced to monovalent copper ions (Cu+) and cleared by thiol compounds inside the cell, causing Zeocin to be activated and able to bind to DNA inside the cell, causing it to break and ultimately leading to cell death.
The protein that produces resistance to Zeocin is derived from the Sh ble gene of Streptococcus hindustanus, which encodes a 14kDa protein that can bind to Zeocin at a certain ratio, preventing it from binding to cellular DNA and inhibiting its DNA fragmentation activity, thereby making cells resistant to Zeocin. Therefore, Zeocin can be used to screen for polyclonal or monoclonal cells expressing Zeocin resistance genes, or for the maintenance culture of corresponding polyclonal or monoclonal cells.
Zeocin is effective for the vast majority of aerobic cells and is commonly used for screening bacteria (such as Escherichia coli), eukaryotic microorganisms (such as yeast), and animal and plant cells. The recommended concentration for screening Escherichia coli is 25-50 μ g/ml (low salt LB medium, NaCl concentration should not exceed 5g/L); The recommended concentration for yeast screening is 50-300ug/ml (YPD or basic medium); The recommended concentration for mammalian cell screening is 50-1000ug/ml (suitable culture medium, depending on the type of cell line). In practical applications, the concentration gradient of Zeocin should be tested for different cell lines to determine the optimal concentration for use.
