Evagreen is a DNA binding dye used for real-time quantitative PCR (qPCR). The many advantages of this dye make it far superior to SYBR Green I. In addition to having similar spectral characteristics, Evagreen has three main features that distinguish it from SYBR Green I:
The inhibitory effect of Evagreen on PCR is much lower than that of SYBR Green I, and qPCR experiments using Evagreen can use rapid PCR steps. Meanwhile, Evagreen can use higher concentrations in the experiment to obtain amplification signals much stronger than SYBR Green I. The higher concentration of Evagreen also eliminates the defect of "dye redistribution", making Evagreen suitable for both multiplex PCR and high-resolution melting curve analysis (HRM). This analysis is increasingly being used for post PCR genotyping and heterodyne analysis. Due to the inhibitory effect of SYBR Green I on PCR, its usage concentration must be very low. Therefore, SYBR Green I cannot solve the problem of dye redistribution caused by low concentration and cannot be used for multiplex PCR or HRM. Meanwhile, the issue of dye redistribution may also affect the reliability of conventional melting curves, as low melting point DNA strands may not be detectable due to this reason.
Evagreen has extremely strong stability and will not be damaged during normal storage, operation, and PCR processes. Dyes in buffer solutions can be safely stored at room temperature or in the refrigerator, and can also be repeatedly frozen and thawed. On the contrary, SYBR Green I is unstable and has stronger PCR inhibition after degradation.
Evagreen reduces cell membrane permeability, making it safer than SYBR Green 1. Independent laboratory tests have shown that Evagreen is neither mutagenic nor cytotoxic. On the contrary, although SYBR Green I itself has weak mutagenicity, it may inhibit the normal DNA repair mechanism in cells, giving it a mutagenic enhancing effect. Considering the widespread use of PCR, its safety should be given sufficient attention.
