G418 Sulfate, also known as Geneticin, G418, G418, or G-418, is an aminoglycoside antibiotic with a structure similar to Gentamycin B1. It originates from Micromonospora rhodorangea and is commonly used to screen eukaryotic or prokaryotic polyclonal or monoclonal cells expressing the neo gene. G418 is not only used for screening stable cell lines, but also for maintaining stable cell lines. The 50mg/ml packaging of this product has been filtered and sterilized, and can be directly used for cell culture.
In bacteria, yeast, protozoa, worms, higher plants, and mammals, genetic mycin inhibits or kills cells by suppressing protein synthesis. Its mechanism of action is that geneticin can bind to 70S and 80S ribosomes, thereby blocking peptide chain extension and interfering with protein synthesis. Its resistance genes (mainly neo genes) are located on transposons Tn601 (903) or Tn5 (derived from bacteria) and can be expressed in eukaryotic cells. By using genetic recombination technology to introduce these resistance genes into cells, the cells express the resistance product aminoglycoside 3 '- phosphotransferase (APH (3) II), thereby obtaining resistance to G418, which is used to screen and maintain prokaryotic or eukaryotic cells carrying resistance genes in culture.
In mammalian cells, when the resistance gene neo is integrated into the eukaryotic genome, it encodes and expresses aminoglycoside 3 '- phosphotransferase (APH (3) II). This enzyme covalently modifies the amino or hydroxyl function of G418, inhibiting the interaction between antibiotics and ribosomes, thereby rendering antibiotics inactive. This characteristic endows cells with resistance and enables them to grow in selective media containing G418. When conducting stable cell line screening experiments, it is necessary to establish a dose-response curve or kill curve to determine the minimum effective concentration for killing non resistant cells.
In plant cells, resistance is incubated by transfecting resistance plasmids carrying the nptII gene. The nptII gene can also encode and express aminoglycoside phosphotransferase, which inactivates various antibiotics including G418, kanamycin, and paromycin.
Both G418 and neomycin are aminoglycoside antibiotics that can be inactivated by Neo gene products, making them suitable for screening cells carrying Neo resistance genes. Cells screened for neomycin can be replaced with G418.
